چکیده

I am honored to introduce the new journal Cells, which has been created to serve as a hub for disseminating new findings and discoveries in cell biology to researchers worldwide. Cells is an international, peer-reviewed, open-access journal on cell biology, molecular biology, and biophysics. Much has been accomplished in molecular and cell biology in the past thirty years. We have unraveled the structure of many proteins, nucleic acids and lipids, and learned much about cell structures, including lysosomes, mitochondria, cytoskeletal elements, cell membranes, endoplasmic reticulum, and nuclei. Now, it is time to apply this knowledge to answering the age-old question: "How do cells work?" It appears there is a strong demand for a journal focused on various functional aspects of cell biology, including such fundamental ones as gametogenesis, embryonic development, tissue regeneration, tumorigenesis, and aging. Multiplexed Elispot assay.

Micron scale latex beads are well established as highly biocompatible reagents. Imbibing two fluorescent dyes into the interior of the beads enables the creation of a family of combinatorially colored labels. Previous use of such beads, in flow cytometry for example, has focused on beads of approximately 5 microm diameter. We show here that 280 nm combinatorially labeled particles can be used t...

Enzyme-Linked Immunosorbent Spot Assay for the Detection of Wilms’ Tumor 1-Specific T Cells Induced by Dendritic Cell Vaccination

BACKGROUND Despite recent advances in cancer immunotherapy and the development of various assays for T cell assessment, a lack of universal standards within immune monitoring remains. The objective of this study was to evaluate the enzyme-linked immunosorbent spot (ELISpot) assay in comparison with major histocompatibility complex-tetramer analysis in the context of dendritic cell (DC)-based ca...

Computer-assisted, quantitative cytokine enzyme-linked immunospot analysis of human immune effector cell function.

Originally developed for detecting antibody production from B lymphocytes, the enzyme-linked immunospot (ELISPOT) assay was later modified to assess cytokine production from various immune effector cells. Although the ELISPOT assay can detect antibody or cytokine production at the single-cell level, the visual counting of spots in a 96-well plate under a microscope makes this method unsuitable ...

Chemistry and biology of the ELISPOT assay.

Enzyme-linked immunospot, or ELISPOT, assay allows the detection of low frequencies of cells secreting various molecules. ELISPOT can be used in many areas of research and, because of its high sensitivity, has the potential to become a valuable diagnostic tool. Based on the same "sandwich" immunochemical principles as enzyme-linked immunosorbent assay, ELISPOT is easy to perform and quantify th...

ELISPOT Assay for Monitoring Cytotoxic T Lymphocytes (CTL) Activity in Cancer Vaccine Clinical Trials

The profiling and monitoring of immune responses are key elements in the evaluation of the efficacy and development of new biotherapies, and a number of assays have been introduced for analyzing various immune parameters before, during, and after immunotherapy. The choice of immune assays for a given clinical trial depends on the known or suggested immunomodulating mechanisms associated with th...
